With a test cord to plug into the ONT.
OK, I guess that measures light power of the ONT, but doesn't take into account the 'loss' of the return light power back to OLT.

I'm assuming that the loss could be different in either direction due to different frequencies and how the light interacts with the prisms in the splitters?

Obviously you can't expect to measure the light received at the OLT, only the output of the ONT.
